==== Blood-cell formation ==== The specificity of the human immune-cell repertoire is what allows the human body to defend itself from rapidly adapting antigens. However, the immune system is vulnerable to degradation upon the pathogenesis of disease, and because of the critical role that it plays in overall defense, its degradation is often fatal to the organism as a whole. Diseases of hematopoietic cells are diagnosed and classified via a subspecialty of pathology known as hematopathology. The specificity of the immune cells is what allows recognition of foreign antigens, causing further challenges in the treatment of immune disease. Identical matches between donor and recipient are no longer required for successful transplantation. Rather, haploidentical matches have facilitated numerous transplants, given improvements in post-transplant immunosuppressive regimens. Research using both hematopoietic adult stem cells and embryonic stem cells has provided insight into the possible mechanisms and methods of treatment for many of these ailments. Fully mature human red blood cells may be generated ex vivo by hematopoietic stem cells (HSCs), which are precursors of red blood cells. == Historical understanding == Historically, there has been much dispute over whether such a thing as a vacuum can exist. Ancient Greek philosophers debated the existence of a vacuum, or void, in the context of atomism, which posited void and atom as the fundamental explanatory elements of physics. Lucretius argued for the existence of vacuum in the first century BC and Hero of Alexandria tried unsuccessfully to create an artificial vacuum in the first century AD. Following Plato, however, even the abstract concept of a featureless void faced considerable skepticism: it could not be apprehended by the senses, it could not, itself, provide additional explanatory power beyond the physical volume with which it was commensurate and, by definition, it was quite literally nothing at all, which cannot rightly be said to exist. Aristotle believed that no void could occur naturally, because the denser surrounding material continuum would immediately fill any incipient rarity that might give rise to a void. In his Physics, book IV, Aristotle offered numerous arguments against the void: for example, that motion through a medium which offered no impediment could continue ad infinitum, there being no reason that something would come to rest anywhere in particular. In all other aspects, however, the RF government perpetuated existing racial segregation and inequalities: the white minority's economic domination and ownership of land was maintained, as was the racial segregation of public services, education and electoral rolls through the party's policy of "separate economic advancement". Combined with a largely successful boycott campaign from the black majority, this resulted in de facto white minority rule. In an exception to their usual policies, the 1969 constitutional reform explicitly delineated the two electoral rolls by race: With the European 'A' roll increased to 50 seats as opposed to the African 'B' roll only having 8 (with an additional 8 indirectly elected to represent chiefs and tribal interests), this resulted in 270,000 whites having 50 seats and 6 million Africans having 16 seats in the Assembly. == Pathophysiology == RA primarily starts as a state of persistent cellular activation leading to autoimmunity and immune complexes in joints and other organs where it manifests. The clinical manifestations of disease are primarily inflammation of the synovial membrane and joint damage, and the fibroblast-like synoviocytes play a key role in these pathogenic processes. === Classical PKU === Classical PKU, and its less severe forms "mild PKU" and "mild hyperphenylalaninemia" are caused by a mutated gene for the enzyme phenylalanine hydroxylase (PAH), which converts phenylalanine to tyrosine, which is a conditionally essential amino acid for PKU patients, because without the enzyme PAH, tyrosine cannot be produced in the body through oxidation of Phe. PAH deficiency causes a spectrum of disorders, including classic phenylketonuria (PKU) and mild hyperphenylalaninemia (also known as "hyperPhe" or "mild HPA"), a less severe accumulation of phenylalanine. Compared to classic PKU patients, patients with "hyperPhe" have greater PAH enzyme activity and are able to tolerate larger amounts of phenylalanine in their diets. Without dietary intervention, mild HPA patients have blood Phe levels higher than those people with normal PAH activity. Currently, no international consensus exists on the definition of mild HPA, but it is most frequently diagnosed at blood Phe levels between 2 and 6 mg/dL. Phenylalanine is a large, neutral (hydrophobic) amino acid (LNAA). LNAAs compete for transport across the blood–brain barrier (BBB) via the large neutral amino acid transporter (LNAAT). If phenylalanine is in excess in the blood, it saturates the transporter. Excessive phenylalanine levels tend to decrease the levels of other LNAAs in the brain. As these amino acids are necessary for protein and neurotransmitter synthesis, Phe buildup disrupts the development of the brain, causing intellectual disability.
